Abstract
Human metaphase chromosomes were dissected using an atomic force microscope (AFM) in ambient conditions and in buffer. Cutting with z-modulation in air yielded precise cuts at loading forces F> 17 μN with a full width at maximum depth of 90 nm. After dissection, the chromosomal material adhered to the tip to be used for further biochemical processing. In liquids, we measured the effects of different types of buffer solution on swelling of the chromosomes and their elastic behaviour.
