Abstract

Basophilic granulocytes are mastocytes which can be generated from patients' whole blood through the use of certain separation methods. These granulocytes can then be used for diagnostic purposes to determine allergic or pseudo-allergic diseases through in-vitro tests (as, e.g., for the determination of an occupational disease). With the isolated granulocytes, it is possible to perform provocation tests on a culture using defined allergen or pseudoallergen concentrations; after these provocation tests, the specifically set free mast cell substances as histamine or tryptase can de determined. Mastocyte tryptase is more sensitive than histamine and can be determined with the help of a spectrometer after it has been set free during the provocation test. Thus, it is possible to avoid expensive and time-consuming provocation and exposure tests on the patient himself.
