Internal motility in stiffening actin-myosin networks

Abstract

The study of actin networks containing the heavy meromysin (HMM) subfragment lacking the light meromysin domain responsible for myosin II assembly was discussed. The main focus of the study was on the viscoelastic behavior and internal dynamics of such networks during ATP depletion. A sol-gel transition accompanied by a sudden onset of directed filament motion was identified with the use of an experimental set up combining microrheology with fluorescent microscopy of labeled filaments. The results show that a zipping mechanism was needed to explain the filament motion in the vicinity of the sol-gel transition.
