N2 - Background: How are isotype-mixed HLA class II heterodimers selected from the large pool of α-β subunit combinations? Results: Two critical sequences in DPΒ and the chaperone invariant chain control assembly of isotype-mixed HLA-DRαDPβ heterodimers. Conclusion: A substantial proportion of HLA-DPβ allotypes, with the DPβ motif Lys-69/GGPM-(84-87), form matched heterodimers with DRα.
