20S proteasome and its inhibitors: Crystallographic knowledge for drug development

Abstract

An overview of the latest insights on the structure and function of the proteasome is given highlighting the structural features that determine its activity and specificity. Focus is on the current structural knowledge of the mechanisms of regulation and modulation of proteasomal activity by various natural and synthetic chemical compounds, showing the proteasome as a potential target for drug development in biomedical research. The mechanisms in action of various proteasomal inhibitors and spans scientific publications from 1997 to 2006 are described in detail.
