An intravenously administered dose of insulin after some 15 minutes is distributed in a volume quantitatively corresponding to the extracellular volume. For certain studies it is important to know the volume in which endogenous insulin is distributed immediately after rapid release, or exogenous insulin immediately after a rapid injection. From the graph of the arterial insulin level in the first 5 minutes after injection of a unit of unlabeled insulin this primary distribution space was therefore determined. This distribution space cannot quantitatively be attributed to any of the usual fluid spaces of the body. Its determination is a prerequisite to calculation of the insulin secretion in the early phase.
